Ten stallions at the Flyinge National Stud were used in the study and three ejaculates were collected from each stallion. The ejaculates were analyzed ten times under different conditions, such as time, storing temperature and with/without selection. The selection was carried out by Single Layer Centrifugation. The method for analysis of the spermatozoa was computerized fluorometry of Hoechst 33258-stained semen samples. The objective of the study was to:
• evaluate the viability with/without selection, at different times and at different storage temperatures.
• evaluate if there is correlation between viability and subjective motility.
The results of the selection experiments were interesting. Selected spermatozoa had a higher motility and viability after storage, especially after 48 h, than the unselected spermatozoa (P<0,0001). The method to evaluate stallion spermatozoa viability by computerized fluorometry showed various results and correlation between viability and motility was over-all significant (r=-0,59, P<0,0001) but not significant under certain conditions. |

Hästaveln är inriktad på prestation vilket medför att det är stor variation på spermakvaliteten och fertiliteten hos de hingstar som används i avel. Den ökade användningen av kyld transportsperma vid artificiell insemination har resulterat i sjunkande dräktighetsresultat. Detta gör att det finns ett behov av att utvärdera och utveckla nya metoder för att kvalitetsbedöma samt kvalitetssäkra hingstsperma avsedd för semin. Denna studie är en del av ett större forskningsprojekt med just detta ändamål. Projektet drivs av SLU med bland annat Flyinge AB som samarbetspartner. Tio hingstar stationerade på Flyinge ingick i studien och från vardera hingst togs tre ejakulat. Ejakulaten analyserades totalt tio gånger vid olika tidpunkter, förvaringstemperatur samt med eller utan selektion. Selektion skedde genom Single Layer Centrifugering. Metoden som användes vid analys av sperman var datoriserad fluorometri med Hoechst 33258 som färgmarkör. Syftet med studien var att • utvärdera spermiernas viabilitet med respektive utan selektion av spermierna, vid olika tidpunkter och vid olika förvaringstemperatur. • utvärdera om det finns någon korrelation mellan viabilitet och subjektiv motilitet. Resultaten i denna studie vad gäller selektionsmetoden är intressanta. Den selekterade sperman har en högre motilitet och viabilitet över tid, framför allt efter 48h, än den oselekterade sperman (P<0,0001). Metoden att analysera viabiliteten hos hingstsperma med hjälp av datoriserad fluorometri är dock tveksam. Resultaten är ojämna och korrelation med motiliteten är totalt sett stark (r=-0,59, P<0,0001) men saknas helt vid vissa tidpunkter och vissa behandlingar.
