description Time lapse microscopy imaging has revolutionized the study of dynamic events taking place inside living cells. When combined with automated image analysis it provides a powerful tool that enables spatiotemporal large-scale quantification at the level of complete cell population. However, existing image analysis methods are rather limited in analyzing such time lapse datasets. Here is presented a fully automated approach to analyze phase contrast and fluorescence microscopy images using CellProfiler. Cellprofiler is an open source modular package that contains advance algorithms for image analysis for extraction of quantitative information from biological images. In this study pipelines based on CellProfiler were designed and implemented for analyzing drug induced apoptosis and proteasome inhibition in cancer therapy. A method for automatic extraction of apoptotic cells using phase contrast images based on their intrinsic shape features is also proposed.
