In this study the avidity (i.e the antigen binding force) of BRSV specific antibodies was measured to see if there was a difference between antibodies produced during an acute phase of infection and antibodies produced by earlier infected animals. A commercially available ELISA-testkit against BRSV-specific antibodies was used and an incubation step with 6M urea was added. The effect of the urea is that it breaks the weak bonds between antibodies and antigen while the stronger bonds remain intact. Four different groups of animals were included in this study; seven calves that were naturally infected, three acutely infected calves with known time of infection, five cows that were seropositive during several years and four experimentally infected calves that had been a part of a vaccine trial. The results of this study showed that antibodies produced during the acute phase of an infection had a low avidity and that the avidity increased with time after infection. Antibodies produced by animals that have been seropositive for a long time and the maternally derived antibodies had a high avidity. There was no correlation between the amount of antibodies and avidity.

Viruset ger stora förluster och dödligheten i en besättning kan vara så hög som 20 %. I den här studien undersöktes aviditeten (den totala bindningskraften mellan en polyklonal antikropp och dess antigen) hos BRSV-specifika antikroppar för att se om det var någon skillnad mellan antikroppar som producerats i en akut fas av infektion och antikroppar producerade av ett djur som varit seropositivt en längre tid. Ett kommersiellt ELISA-testkit mot BRSV-specifika antikroppar användes och en tvätt med 6M urea lades till. Principen är att urean bryter de svaga bindningarna mellan antigen och antikropp medan de starka bindningarna förblir opåverkade. Fyra olika djurgrupper undersöktes; sju kalvar som var naturligt infekterade, tre akut infekterade kalvar med känd infektionstid, fem kor som var seropositiva under en längre tid och fyra experimentellt infekterade kalvar som ingått i en vaccinstudie. Resultaten som sågs i denna studie var att antikropparna som producerades i en akut fas av infektionen hade en låg aviditet och att aviditeten steg ju längre tid som gått från infektionstillfället. Antikroppar hos djur som varit seropositiva under en lång tid samt antikroppar av maternellt ursprung hade hög aviditet.
