The increased use of transport semen requires new methods to ensure good semen quality. In this part of the project the viability and concentration in fresh and cooled semen was examined with NucleoCounter SP-100. A comparison between two different commercially available semen extenders, Kenney's and INRA 96, was made. The selection method Single Layer Centrifugation (SLC) was used in the study and the spermatozoal viability was compared between the selected and unselected semen. Since the NucleoCounter SP-100 is a relatively new machine a comparison was also made with flowcytometry regarding viability. Concentration measured with NucleoCounter SP-100 was compared by the use of manual counting with the Bürker chamber. The results show that no statistical difference was detected between the two different extenders regarding spermatozoal viability. On the other hand SLC had a statistically positive effect on the viability of the spermatozoa over time (p < 0,0001). A significant interaction was noticed between semen extender (Kenney's och INRA 96) and handling of semen (unselected and selected). Viability was best conserved in Kenney's selected and INRA 96 selected had the next best ability to conserve viability over time. Kenney's unselected had the lowest result of viability over time. The comparison between NucleoCounter SP-100 and flowcytometry regarding spermatozoal viability didn't show any correlation. In this part of the study only 19 ejaculates were included and a greater number of stallions and ejaculates should be used for more accurate results. Concerning the concentration measurement, the NucleoCounter SP-100 correlated very well with Bürker chamber (r = 0,93) and the significance was high (p < 0,0001). The results show that SLC has a good effect on sperm viability over time while choice of semen extender does not have any significant effect. NucleoCounter SP-100 could be used at the studs for concentration measurement but regarding viability, a larger study should be made to evaluate the NucleoCounter machine.

Denna studie är en del av ett större projekt vars syfte har varit att undersöka metoder för att kunna kvalitetssäkra hingstsperma. Orsaken till detta är att en del hingstar som används inom hästaveln har nedsatt spermakvalitet. I och med den ökade användning av transportsperma krävs nya metoder för att kunna säkerställa spermans kvalitet. I detta delprojekt undersöktes viabiliteten och koncentrationen hos färsk och kyld sperma med NucleoCounter SP-100. En jämförelse av två olika kommersiellt tillgängliga spädningsvätskor, Kenney’s och INRA 96, gjordes. Selektionsmetoden Single Layer Centrifugation (SLC) innefattades i studien och viabilitet jämfördes i de selekterade ejakulaten med de oselekterade. Eftersom NucleoCounter SP-100 är en relativt ny apparat gjordes en jämförelse mellan denna och flödescytometri beträffande viabilitet. Koncentration uppmätt med NucleoCounter SP-100 jämfördes med Bürkerkammar-metoden. Resultaten visade att det inte förelåg någon statistisk skillnad i hur väl viabiliteten bevarades i de olika spädningsvätskorna. Däremot hade SLC en statistiskt positiv effekt på spermiernas viabilitet över tid (p < 0,0001). Ett signifikant samspel förelåg mellan spädningsvätska (Kenney’s och INRA 96) och behandling (oselekterad och selekterad). Viabiliteten bevarades bäst i Kenney’s selekterade och INRA 96 selekterade hade näst bäst förmåga att bevara viabilitet över tid. Sämst förmåga hade Kenney’s oselekterade. Jämförelsen av NucleoCounter SP-100 med flödescytometri vad gäller spermieviabilitet visade ingen korrelation. I denna del av studien jämfördes enbart 19 ejakulat och ett större provmaterial behövs för att kunna få tillförlitliga resultat. Beträffande koncentrationsmätning korrelerade NucleoCounter SP-100 mycket väl med Bürkerkammare ( r = 0,93) och signifikansen var hög (p < 0,0001). Resultaten visar att SLC har god effekt på spermiernas viabilitet över tid, medan val av spädningsvätska inte har någon signifikant effekt.
