In this study we used an effective method to prevent platelet aggregation. Blood was collected in CTAD test tubes and prostaglandin E1 was added to the blood sample. Blood samples were collected from nine healthy cats 1-10 years of age. The blood was analysed within 1-4 hours at the laboratory of clinical chemistry. The platelets were counted both manually and in automated cell counters. Three cell counters were used. A Sysmex XT 2000Vi counted platelets both optically with a laser system and by using impedance analysis. The Siemens Advia 2120 used an optical laser system to count platelets. The Cell-Dyn 3500 counted platelets by impedance. The blood samples were checked for platelet aggregation in the hemocytometer (glass chamber) and on the blood smears. Platelet aggregation was prevented when adding prostaglandin E1 to blood samples directly after they were collected in CTAD test tubes. Since no or only a few small platelet aggregates were present in the blood samples after addition of prostaglandin E1 to the CTAD test tubes, the platelet counts aquired from the nine cats can be used as reference values for the instruments and manual method used in this study. The reference values in this study were 180-493x109/L for Sysmex XT 2000Vi optical analysis, 182-525 x109/L for Siemens Advia 2120 and 161-380 x109/L for the manual platelet counts. Moreover the CTAD-prostaglandin E1 method may be used with feline patients to obtain accurate platelet counts and allow better diagnosis.

Trombocytaggregering vid blodprovstagning på katt är ett stort problem vid trombocyträkning. Svårigheterna vid analys av trombocytantal hos katter har medfört att inga tillförlitliga referensvärden för trombocytantal finns i dagsläget. Vi har i denna studie tagit fram en ny metod för att förhindra trombocytaggregeringen. Blodprov togs på nio stycken friska katter i åldrarna 1- 10 år i CTAD-rör. Till blodproven tillsattes prostaglandin E1 i samband med blodprovstagningen. Blodproven analyserades inom 1-4 timmar på laboratoriet för klinisk kemi och trombocyterna räknades både manuellt och med automatiserade analysatorer. Analysatorerna som användes var Sysmex XT 2000Vi som räknade trombocyterna både optiskt och med impedansmätning, Siemens Advia 2120 som är en optisk cellräknare och Cell-Dyn 3500 som är en impedanscellräknare. Vid den manuella räkningen kontrollerades även förekomsten av trombocytaggregat i blodproven. Resultaten visar att trombocytaggregering förhindras vid tillsats av prostaglandin E1 till blodprover tagna i CTAD-rör. Eftersom inga eller endast enstaka små trombocytaggregat sågs när prostaglandin E1 tillsattes till CTAD-rör kan trombocytvärdena från de nio katterna användas som referensvärden för de metoder och de instrument som användes i den här studien. Referensvärdena blev 180-493x109/L för Sysmex XT 2000Vi optisk analys, 182-525 x109/L för Siemens Advia 2120 och 161-380 x109/L för den manuella räkningen.
