description Melilotus albus is a legume plant naturalized in Argentina with a great potential to be used as a forage resource in restricted environments. Our objective was to characterize 10 accessions of M. albus belonging to a collection from INTA and from the FCA-UNL by using molecular markers. Five ISSR primers producing 52 bands, 90 % of them polymorphic, were found to be equally informative. Eight SSR loci were amplified generating 30 alleles, averaging 3.8 alleles per locus, almost all of them highly informative. In numerical analysis, separation among accessions was unclear; genotypes congregate in complex intermixed groups. The greatest proportion of the variation was registered within accessions, in accordance with the allogamous reproductive nature of the species. Since neutral markers were used, the high variability encountered within accessions could be related to gene flow among them and/or a similar origin of the introduced materials. Results from this study support the fact that ISSR and SSR markers provide complementary information and that they are valuable and effective tools to be used to characterize the available germplasm of the species, although other markers such as the functional ones are recommended to better understand the variability in the species and its use in breeding programs.

Melilotus albus es una leguminosa naturalizada en Argentina con gran potencial como recurso forrajero para ser utilizado en ambientes marginales. El objetivo de este trabajo fue caracterizar 10 introducciones de la colección de INTA y de la FCA-UNL mediante marcadores moleculares. Cinco primers ISSR produjeron 52 bandas, 90 % de ellas fueron polimórficas, resultando todos los primers igualmente informativos. Los 8 loci SSR amplificados generaron 30 alelos, con un promedio de 3,8 alelos por locus, siendo altamente informativos la gran mayoría de ellos. En los análisis mediante técnicas numéricas la separación entre grupos no fue clara; los genotipos se congregan en grupos entremezclados. La mayor proporción de la variación se observó dentro de las introducciones, correspondiendo con el modo de reproducción alógama de la especie. Por tratarse de marcadores moleculares neutrales, la elevada variabilidad común entre introducciones podría explicarse por la existencia de considerable flujo génico entre ellas y/o por un origen similar de los materiales introducidos. Los resultados de este estudio demostraron que los marcadores moleculares ISSR y SSR brindan información complementaria y son herramientas valiosas y eficaces para caracterizar el germoplasma disponible de esta especie, pero se precisa incluir a otros marcadores como los funcionales con el fin de tener una mejor comprensión de la variabilidad disponible y su aprovechamiento en programas de mejoramiento genético.
