description The main passionfruit production areas in 11 departments of Colombia, were surveyed and a total of 338 samples collected from diseased passionfruit plants. Samples were examined by electron microscopy, serology, ADNc amplification, cloning and sequencing of viral nucleic acids. The survey and analyses conducted, demonstrated the existence of two different viruses . The prevalent viral pathogen in all departments surveyed, was a species of the Potyvirus genus. An isolate of this potyvirus from Roldanillo, Valle, was cloned and partially sequenced, demonstrating a high nucleotide sequence identity (97%) with Soybean mosaic virus (SMV). The second virus, detected only in the departments of Valle, Antioquia and Santander, was shown to be a distinct species of the Tymovirus genus (60-76% homology with tymoviruses characterized at the molecular level). However, this virus was antigenically related to Passion fruit yellow mosaic virus, a tymovirus that induces similar symptoms in passionfruit in Brazil. Consequently, this name will be retained for the Colombian tymovirus until an isolate of the Brazilian tymovirus is characterized at the molecular level. The possibility of producing virus-free planting material of commercial passionfruit varieties, coincides with the need to increase passionfruit production in Colombia in order to meet the increasing internal and external demand for these fruits and their products.

Se visitaron 11 departamentos productores de maracuyá (Passiflora edulis) en Colombia, donde se colectaron 338 muestras de plantas del frutal aparentemente afectadas por enfermedades virales. El análisis de estas muestras se realizó mediante técnicas de patogenicidad, microscopia electrónica, serología con anticuerpos monoclonales y policlonales, amplificación dc ADNc, clonación, hibridación y secuenciación de ácidos nucléicos. Se detectaron dos virus, siendo el más prevalente (incidencia > 95%) un potyvirus transmitido por áfidos. Un aislamiento de este potyvirus procedente de Roldanillo, Valle, se amplificó mediante la técnica molecular de RT-PCR, se clonó y secuenció, estableciendose una relación de identidad superior al 97% con aislamientos del Virus del mosaico de la soya (SMV). Este virus causa también la enfermedad de la hoja morada de la granadilla (Passiflora ligularis) e infecta P. quadrangularis (badea), P. adenopoda, P. caerulea, y P. pinnatistipula (chulupa). El segundo virus detectado en los departamentos del Valle, Antioquia y Santander, se identificó como una especie del género Tymovirus. Este virus es diferente a otros tymovirus caracterizados a nivel molecular (60-76% de identidad en la secuencia del genoma parcial secuenciado), pero mostró ser patogénica y antigenicamente similar al Virus del mosaico amarillo del maracuyá descrito en Brasil. Por consiguiente, se conserva este nombre para el tymovirus colombiano mientras se caracteriza un aislamiento brasileño a nivel molecular. El tymovirus también infectó P. adenopodo, P. pinnatistipula, and P. quadrangularis. La producción de material reproductivo de maracuyá, granadilla, badea y otras pasifloras libre de virus, coincide con un interés creciente por fomentar el cultivo y así suplir la creciente demanda interna y externa de pasifloras en Colombia. 2001 2014-10-02T08:33:30Z 2014-10-02T08:33:30Z Journal Article https://hdl.handle.net/10568/44256 es Open Access application/pdf
