description A collection of 45 isolates of Sphaerotheca pannosa var. rosae was established from rose plants infected with powdery mildew, one of the diseases caused by this pathogen. After a preliminary molecular screening using RAPD, 16 isolates, collected in floricultural enterprises located in five municipalities of the savannas of Bogotá, Colombia, were seleced as representative of the variability present in S. pannosa. A practical methodology to conserve this obligated pathogen was developed using gelatin capsules, thus providing a permanent source of inoculum for pathological and molecular analyses. Pathogenicity studies conducted with rose cultivars Aalsmeer Gold, Charlotte, Classy, Konfetti, Livia, and Tineke showed that isolate Sp6 did not sporulate in cultivar Tineke, although this isolate affected all other cultivars. The other isolates sporulated on all cultivars, suggesting the presence of at least two pathotypes in the target population. A pathogen-host plant interaction therefore exists. The Internal Transcribed Space (ITS) of the 5.8S gene of ribosomal DNA was amplified using PCR, and a homogeneous and reproducible band, with a molecular weight of 295 bp, was obtained. The amplified product was digested with restriction enzymes that showed a similar band pattern for all isolates, indicating that the isolates belonged to the same species. Intraspecific polymorphisms were detected with 11 RAPD primers and a RAMS microsatellite primer. Three genetic groups, with 60% similarity (Dice index), were detected, indicating intraspecific genetic diversity. The high correlation (+0.89) between origin and genetic diversity, and between this and pathogenic variation (+0.63), suggests that differences exist in environment conditions and/or disease management among farms or in rose cultivars used.

Se estableció una colección de 45 aislamientos de Sphaerotheca pannosa var. rosae a partir de plantas de rosa afectadas con mildeo polvoso, enfermedad causada por este patógeno. Para efectos de este estudio, se hizo una evaluación molecular previa mediante RAPD a todos los aislamientos y se seleccionaron 16 aislamientos contrastantes, como representativos de la variabilidad presente en S. pannosa, los cuales provenían de fincas de floricultores, ubicadas en cinco municipios de la Sabana de Bogotá. Se desarrolló una metodología práctica para la conservación en cápsulas de gelatina, de este patógeno obligado, lo que permitió tener fuente de inóculo permanente para los análisis patológicos y moleculares que se realizaron. Los estudios de patogenicidad en los cultivares Aalsmeer Gold, Charlotte, Classy, Konfetti, Livia y Tineke demostraron que en este último no había esporulación del aislamiento Sp6, pero este aislamiento si causó enfermedad en los otros cultivares. Por su parte, el resto de aislamientos, esporularon en todos los cultivares, lo que sugiere la presencia de al menos dos patotipos en la población analizada lo que sugiere una interacción patógeno-hospedante. Mediante PCR se amplificó la región del Espacio Interno Trascrito (ITS) del gen 5.8S del ADN ribosomal y se obtuvo una banda homogénea y reproducible de un peso molecular de 295 pb. El producto amplificado fue digerido con enzimas de restricción, las cuales mostraron un patrón de bandas igual para todos los aislamientos con cada enzima, lo que indica que todos los aislamientos corresponden a una sola especie. Se pudieron detectar polimorfismos intraespecíficos con 11 cebadores RAPD y un cebador microsatélite RAMS, los cuales permitieron la identificación de tres grupos genéticos a un 60% de similitud (Índice de Dice), estableciéndose que existe variación genética dentro de la especie.
