Accumulation of transgene-derived siRNAs is not sufficient for RNAi-mediated protection against Citrus tristeza virus in transgenic Mexican lime
Publicado 2017“…P>Mexican lime plants transformed with the 3'-terminal 549 nucleotides of the Citrus tristeza virus (CTV) genome in sense, antisense and intron-hairpin formats were analysed for transgene-derived transcript and short interfering RNA (siRNA) accumulation, and for CTV resistance. …”

Post-transcriptional gene silencing of the p23 silencing suppressor of Citrus tristeza virus confers resistance to the virus in transgenic Mexican lime
Publicado 2017“…Previously, we have shown that most Mexican limes (Citrus aurantifolia (Christ.) Swing.) expressing the p23 gene of Citrus tristeza virus (CTV) exhibit aberrations resembling viral leaf symptoms. …”

Virus-Viroid Interactions: Citrus Tristeza Virus Enhances the Accumulation of Citrus Dwarfing Viroid in Mexican Lime via Virus-Encoded Silencing Suppressors
Publicado 2017“…An assay to identify interactions between Citrus dwarfing viroid (CDVd) and Citrus tristeza virus (CTV) showed that viroid titer was enhanced by the coinfecting CTV in Mexican lime but not in etrog citron. Since CTV encodes three RNA silencing suppressors (RSSs), p23, p20 and p25, an assay using transgenic Mexican limes expressing each RSS revealed that p23 and, to a lesser extent, p25 recapitulated the effect observed with coinfections of CTV and CDVd.…”

Origin of C. latifolia and C. aurantiifolia triploid limes: the preferential disomic inheritance of doubled-diploid 'Mexican' lime is consistent with an interploid hybridization hypothesis.
Publicado 2018“…Methods: A population of 85 tetraploid hybrids was established between a doubled-diploid clementine and a doubled-diploid 'Mexican' lime and used to infer the genotypes of 'Mexican' lime diploid gametes. …”

RNA Interference against the Three Citrus Tristeza Virus Genes Encoding Silencing Suppressors Confers Complete Resistance to the Virus in Transgenic Mexican Lime Plants
Publicado 2017“…Considering the important role of p23, p20 and p25 in CTV pathogenesis, we obtained transgenic Mexican lime plants with an intron-hairpin vector carrying full untranslatable versions of genes p25, p20, p23 and the 3'-UTR from the CTV strain T36, to attempt silencing their expression in CTV-infected cells. …”

Transformation of Mexican lime with an intron-hairpin construct expressing untranslatable versions of the genes coding for the three silencing suppressors of Citrus tristeza virus confers complete resistance to the virus
Publicado 2017“…Here, we transformed Mexican lime with an intron-hairpin vector carrying full-length, untranslatable versions of the genes p25, p20 and p23 from CTV strain T36 to silence the expression of these critical genes in CTV-infected cells. …”

Constitutive Expression of Untranslatable Versions of the p25 Coat Protein Gene of Citrus tristeza virus (CTV) in Transgenic Mexican Lime Plants Does Not Confer Resistance to the Virus
Publicado 2021“…To develop RNA-mediated resistance (RMR) against Citrus tristeza virus (CTV), Mexican lime plants were transformed with two modified versions of CTV p25 gene: I) the complete p25 ORF derived from the severe strain T305, mutated to make its mRNA untranslatable, and II) an untranslatable fragment of the p25 gene fused to the green fluorescent protein (gfp) gene. …”
